Fbx15

Fbx15 ( Sheet F- box protein 15) is a protein that is used as a marker protein for the isolation of embryonic stem cells from induced pluripotent stem cells ( iPS cells).

Properties

Fbx15 used in the ubiquitin -proteasome system of the substrate-recognition as a component of the protein complex of the E3 ubiquitin ligase SCF - type ( SKP1 - CUL1 -F - box protein ). Fbx15 binds to Skp1 and Cul1.

Fbx15 encoded by the gene FBXO15, which is also used as a marker gene.

In the method for generating iPS cells, somatic cells are reprogrammed in vitro, so that they regain pluripotency and resemble embryonic stem cells. Since not all the cells are reprogrammed successfully reprogrammed cells must be separated from the other cells.

" A introduced into these somatic cell selection system on the basis of an independent ES cell-specific gene ( Fbx15 ) should make it possible to isolate successfully reprogrammed cells. In this way it could be demonstrated that the combined over-expression of four genes - the Oct4, Sox2, Klf4 and c-Myc gene - is sufficient in connective tissue cells to generate ES cell-like cells.
