Lysogeny broth

LB ( lysogeny broth engl. ) is a complex nutrient medium for culturing bacteria ( bacterial culture ). The composition was formulated in 1951 by Giuseppe Bertani. LB is often mistakenly referred to as Luria broth, Lennox broth, or Luria -Bertani medium. LB is the most commonly used medium for the cultivation of Escherichia coli in the microbiology.

Composition

LB is a complex medium, which is now prepared using the following ingredients:

  • Yeast extract (5 g / l)
  • Tryptone (10 g / l)
  • Sodium chloride ( 0.5-10 g / L)

The pH is adjusted to 7. For agar plate cultures (15 g / l ) was added. The medium is autoclaved for 20 minutes at 121 ° C.

In the original procedure 1951, 1 g / l glucose are also included.

The formulations differ in the salt concentration, which are set according to the osmotic conditions of the bacterial strain and desired culture conditions. The low salt formulations, Lennox and Luria, are ideal for cultures requiring salt - sensitive antibiotics such as Zeocin.
