Malonyl -coenzyme A ( shortly: malonyl -CoA) is a thioester of malonic acid with coenzyme A. malonyl -CoA is the molecular mechanism of fatty acid synthesis is required. There take a central role as a Fettsäurekettenverlängerer, also in the biosynthesis of polyunsaturated fatty acids (e.g., arachidonic acid from linoleic acid). Here it provides to its carbon chain fatty acid proportion for extension (and thus creation of double bonds). For the production of malonyl- CoA is a biotin - dependent enzyme which requires acetyl -CoA carboxylase (ACC). Thus, malonyl-CoA is produced by the carboxylation of acetyl -CoA ( activated acetic acid ) as a precursor to the formation of fatty acids. The ACC is the pacemaker enzyme of fatty acid biosynthesis. The incorporation of CO2 proceeds with a simple ATP consumption.

Fatty acid biosynthesis takes place in higher organisms mainly in the liver.
