Propidium iodide ( PI) acts as ethidium bromide as Nukleinsäureinterkalator. The dye may be the perforated cell membranes of dead cells but do not penetrate the intact membrane of living cells. This property is used in flow cytometry for discrimination of cell viability. Another PI flow cytometry is the measurement of the DNA content of cells dar. This is mainly used for analysis of the cell cycle, or for the determination of polyploidy used.

Propidium iodide having an absorption maximum at 488 nm and an emission maximum at 590 nm These maxima shift to 535 nm or 617 nm when intercalated into DNA propidium iodide.
