Protein-Engineering

Protein engineering ( engineering of English here as " construction ", " manipulation " ) is a branch of biochemistry and biotechnology, which deals with the design, optimization and production of proteins, including enzymes, busy. There are basically two approaches in protein engineering: Rational design of proteins ( protein design ) and directed evolution. In parallel, the vector in the course of a vector designs is usually adjusted, where is the gene for the desired protein (transgene ).

Protein Engineering takes place in the construction of Zinkfingernukleasen eg.

Rational Design

The protein design amino acids or whole sequences by targeted, site-specific mutagenesis of the DNA to change from individual properties of the resulting transcribed and translated proteins are exchanged, deleted or inserted.

Directed Evolution

Directed evolution is carried out via random mutagenesis or recombination ( DNA shuffling ), and subsequent selection of the mutants or of the mutant proteins on the desired properties. This approach requires a high-throughput screening.
