SYBR Green I (SG) is an asymmetric cyanine dye that is used in molecular biology for the detection of double stranded DNA.

Properties

SYBR Green I binds to double stranded DNA. The resulting DNA - fluorescent dye complex absorbs blue light at a wavelength lambda max = 494 nm and emits green light at lambda max = 521 nm Further, although clearly weaker, the absorption maxima in the UV range at 284 nm and 382 nm SYBR Green I also binds to single-stranded DNA and RNA. This fluorescence signal is significantly less than when the dye is bound to a double-stranded DNA. SYBR Green I is mutagenic and potentially carcinogenic.

Use

SYBR Green I, is used for detection of double stranded DNA, for example, in gel electrophoresis of real time quantitative PCR and Zellviabilitätstests. Alternatively, ethidium bromide, methylene blue, or silver stain for staining of DNA is used. Similar cyanine dyes for staining of DNA are eg SYBR Green II ( for the detection of RNA and single-stranded DNA ), SYBR Gold, SYBR Safe, YO ( oxazoles Yellow), TO ( Thiazole Orange ) and PG ( PicoGreen ).
