D-6 -phosphogluconate is an intermediate product of carbohydrate metabolism. It is derived from D- gluconic acid, wherein there is esterified on its carbon atom C 6 with phosphoric acid. D-6 -phosphogluconate occurs in the pentose phosphate pathway and the Entner- Doudoroff pathway. The enantiomer L-6 -phosphogluconate does not occur in nature.

The formation of D -6- phosphogluconate in the above-mentioned metabolic pathways runs the same: In both cases it is produced via an intermediate step of D-glucose -6-phosphate, as evidenced by two enzymes, glucose- 6-phosphate dehydrogenase and a Phosphogluconatdehydratase, enzymatically is catalyzed. In this oxidation, NADPH is formed.
