6- phosphogluconolactone is a chemical compound, which occurs as an intermediate in the pentose phosphate pathway. In the first step of the pentose phosphate pathway by the enzyme glucose -6-phosphate dehydrogenase ( G6PDH ) catalyzes the oxidation of glucose -6-phosphate into 6- phosphogluconolactone. Cofactor of the enzyme is NADP , which is to NADPH / H reduced:
Since in this reaction in the form of NADPH Reduktionsäquvialente are to be generated, which are required in other metabolic synthetic pathways, such as the fatty acid, the glucose-6- phosphate dehydrogenase, is specifically responsible for the conversion of NADP to NADPH. NAD can not be used as a cofactor why.

Glucose -6-phosphate is oxidised at the C1 - atom by means of co-factor NADP . In the next step of the pentose phosphate pathway, 6- phosphogluconolactone is hydrolyzed to 6- phosphogluconate.
