Etoposide is a glycoside of podophyllotoxin that from the root of the evergreen American Maiapfels ( Podophyllum peltatum ) is obtained.

Pharmacology

Etoposide is chemically unchanged or in the form of its phosphoric acid ester ( etoposide phosphate ) used as a cytostatic agent in the chemotherapy. Inhibiting the enzyme topoisomerase II, and suppresses the cytokinesis. This means that the cell enters the stage of apoptosis, and dies. Further etoposide leads to single and double -strand breaks in DNA, which enhanced the cytotoxicity.

As side effects of chemotherapy occur predominantly allergies, peripheral neuropathy, hypotension, bronchospasm and mild liver dysfunction.

A very similar compound is the semi-synthetic teniposide, which can be prepared from podophyllotoxin.
