Folylpolyglutamate synthase

The Folylpolyglutamat synthase ( FPGS ) is an enzyme, ligated to the ATP - dependent L- glutamate residues of reduced folic acid derivatives.

Just as polyglutamate these substances can be stored in the cell ( in the cytosol and mitochondria ), and only in this form that they take as a cofactor in several important metabolic reactions in part. The optimum number, in which the cofactors are the most effective reactants in humans six glutamate molecules. FPGS is part of almost all living things.

As the substrate as folic acid antagonists are accepted by FPGS. Resistance of cancer cells to these drugs may partly have mutations in the FPGS gene with the following enzyme deficiency as the cause, since the substances are transported out again unchanged from the cell. Also, inhibitors of FPGS even be used in chemotherapy.
