Fura- 2-AM is a membrane- course derivative of Fura -2. Fura -2 was in 1985 by Roger Tsien and colleagues and serves as a Ca2 indicator.

Properties

Fura-2 is present as a sodium or potassium salt. By the binding of acetoxymethyl ester (AM) can Fura-2 penetrate by passive diffusion into the cell. In the cell, esterases cleave the AM from endogenous, whereby firstly Fura is reactivated, and secondly, leakage from the cell is prevented.

Measurement of Ca2 -induced fluorescence change occurs at 340 and 380 nm, with Ca2 -saturated fura-2 is excited at 340 nm and free Fura -2 at 380 nm. The released amount of calcium is determined by the ratio 340/380 nm, since differences in the distribution of Fura -2 within the cell and differences in cell thickness already be accounted for by this relationship.
