Gal4/UAS-System

Using the GAL4/UAS-Systems (including the UAS -GAL4 system), any cloned genes are expressed selectively in specific cells or tissues. GAL4 is a hefespezifischer transcription factor is under the control of a weak promoter, and codes for the protein GAL4. For the expression of an additional promoter or enhancer is required. GAL4 binds specifically to the protein, the so-called UAS (upstream activating sequence ), whereby a downstream located target gene is activated. This may be, for example, the gene for the green fluorescent protein GFP, or a gene to be examined.

Developed the GAL4/UAS-System by Andrea Brand and Norbert Perrimon at Harvard Medical School for the model organism Drosophila melanogaster and published in Development for the first time in 1993. It is often used in conjunction with molecular-biological methods, such as the enhancer trap method.
