Mestranol is a synthetic drug from the group of estrogens. It is a prodrug ( an inactive precursor) of ethinyl estradiol commonly used in drugs for contraception.

Pharmacological properties

Chemically, mestranol of the 3 - methyl ether of ethinyl estradiol. For the interaction with the estrogen receptor, however, the free hydroxy group is essential, therefore, mestranol is itself inactive. After oral administration of mestranol is absorbed in the gastrointestinal tract and metabolized in the liver by demethylation in ethinylestradiol. The extent of this conversion is highly variable, so mestranol is now used only rarely.

Clinical information

Just as ethinyl estradiol, mestranol can be used for hormone replacement therapy as estrogenic component in combined oral contraceptives (birth control pills), or in preparations. The contraindications ( contraindications), adverse effects and drug interactions are similar to those of ethinyl estradiol.

Mestranolhaltige finished drugs are no longer used in most countries.
