The protein NOD2 is a receptor for foreign material in the innate immune system of mammals, a so-called Pattern - Recognition Receptor. NOD2 is mainly produced by monocytes, specifically recognizes bacterial muramyl dipeptide and starts after detecting the foreign substances on RICK and the IKK -gamma NF kappa B signaling pathway. Mutations in the NOD2 gene are responsible for the rare hereditary Blau syndrome and early-onset sarcoidosis (EOS), as well as of developing for increased risk of developing ulcerative colitis or Crohn's disease.

Recent findings suggest that NOD2 recognizes besides bacterial and viral patterns and activate as a result of over MAVS and IRF3 viral immune response. Together with NOD1 NOD2 is thus a central component of the innate immune system.
