Piroxicam is a drug, specifically a nonsteroidal anti-inflammatory ( NSAID NSAID) from the group of Oxicams with a long half -life ( approximately 50 hours in humans). It is used primarily for pain conditions of the musculoskeletal system (arthritis, rheumatism ).

Mechanism of action

Like almost all NSAIDs piroxicam inhibits reversibly the enzyme cyclooxygenase ( COX). In addition, piroxicam inhibits the migration of monocytes and polymorphonuclear leukocytes, the release of oxygen radicals from activated leukocytes and knorpeldestruierenden of lysosomal enzymes. According to in vitro studies of cartilage metabolism remains unaffected. It works by analgesic ( analgesic ), anti-inflammatory ( anti-inflammatory ) and antipyretic ( antipyretic ).

Piroxicam has the house dog also antitumor activity in certain tumors ( transitional cell carcinoma of the urinary bladder, squamous cell carcinoma of the oral cavity, tubulopapilläre polyps of the rectum ). The exact mechanism of action is unknown, it is presumably due to a reduction in the immunosuppressive effect of prostaglandin E2.

Side effects and contraindications

Other side effects may occur: rash, stomach and duodenal ulcers, edema, hepatitis, cholestasis, colitis, nephritis, malaise, Phototoxie, Henoch-Schönlein purpura, hair loss and high blood pressure. With long-term use, monitoring of renal and liver function tests is necessary.

When blood formation disorders and gastro -intestinal ulcers Piroxicam is contraindicated. During lactation and pregnancy, this product should not be used.

The European Medicines Agency has recommended restricting the use of piroxicam in July 2007 as it comes to more gastrointestinal and skin side effects than other NSAIDs.
