Super Optimal Broth

SOB (Super Optimal Broth ) is a complex nutrient medium which is used primarily for the production and transformation of competent bacteria. The composition was formulated in 1983 by Douglas Hanahan.

Composition

SOB is a complex medium with the following ingredients:

  • Yeast extract 0.5% ( w / v) (5 g / l)
  • Tryptone 2% ( w / v) (20 g / l)

The medium is prepared without the Mg2 and 20 minutes autoclaving at 121 ° C. The nutrient medium is 20 mM (10 ml / l ) was added a 2 M Mg2 solution before use, consisting of 1 M MgCl2 · 6H2O and MgSO4 · 7H2O 1 M was prepared. The pH is 6.8 to 7
