ATP citrate lyase
- OMIM: 108728
- UniProt: P53396
ATP citrate lyase (also ATP-citrate synthase) is the enzyme which transfers an acetyl group of citrate in the cytosol of eukaryotic cells to coenzyme A, as a result, inter alia, acetyl -CoA. This is the second reaction step in the citrate shuttle which is necessary to the herauszubefördern produced in glycolysis acetyl- CoA from the mitochondria. These processes are part of the energy metabolism in all eukaryotes.
The generated acetyl -CoA is the starting material for which takes place in the cytosol lipogenesis.
Hypocitraturie is a cause of kidney stones and may have an excessive activity of ATP - citrate lyase in urine leukocytes as a cause, as determined in a Thai study. Change in diet or potassium supplementation are potential therapies. ATP citrate lyase is a potential target for combating cancer.
Catalyzed reaction
CoA -SH ATP ⇒ ⇒ ADP Pi
Acetyl is transferred under ATP consumption of citrate on CoA: it arises oxaloacetate and acetyl -CoA.
