5 -fluorocytosine (often 5 - flucytosine, trade name: Ancotil ) is a heterocyclic organic compound with a Pyrimidingrundgerüst, an amino group at position 4, a keto group at position 2 and fluorine in position 5, it is a derivative of the nucleobase cytosine. It is used as a systemic antifungal agent for the treatment of mycoses.
Pharmacology
5 -fluorocytosine is regarded as so-called prodrug. By a cytosine permease, it is added to the fungal cell and immediately deaminated by the cytosine deaminase - 5 -fluorouracil. Deamination of 5 -fluorocytosine to 5-fluorouracil is not done in the mammalian cells or only to a small extent. Whether 5 -fluorocytosine in humans may act as a carcinogen, is not yet known.
5 -fluorouracil inhibits DNA synthesis and can lead to the formation of defective RNA, thus it acts fungistatic ( inhibiting fungal growth ). 5 -fluorocytosine is effective against Candida species ( here there is already partially resistance ), Aspergillus species and other fungi. To prevent further development of resistance, it is often combined with amphotericin B.
