Hyperforin is one of the main ingredients of Real St. John's wort. In the plant itself Hyperforin accumulates mainly in fruit punch and where it probably as secondary plant substance has defensive duties. Medical hyperforin is effective, one of the main effects of hyperforin is as an antidepressant. Hyperforin is a reuptake inhibitor of serotonin, dopamine, norepinephrine, GABA and glutamic acids. It inhibits the reuptake of these neurotransmitters from the synaptic cleft. Hyperforin also binds to the enzymes cytochrome P450, CYP3A4 and CYP2C9 and to the pregnane X receptor ( PXR ).

Chemical- structural can be derived by hyperforin Polyprenylierung from phloroglucinol. This latter loses but both its aromaticity as well as the phenolic character. Biochemically created the basic structure of hyperforin from isobutyryl -CoA and three molecules of malonyl -CoA using the isobutyrophenone synthase.
