Altretamine is the generic name of a cytostatic drug (trade name: Hexalen ® ( USA) ), which is chemically is hexamethylmelamine. Altretamine is a six- methylated derivative of melamine and is used as a drug in the treatment of ovarian cancer.

Properties and use as a medicament

Altretamine ovarian cancer ( ovarian cancer) is for the palliative treatment of patients with relapsed ( recurring) or refractory (not responsive ) allowed.

The exact mechanism of action of Altretamine is still largely unknown. It is believed that the DNA - and RNA - synthesis is inhibited in the cell nucleus or that the N -demethylation of altretamine reactive intermediate products which bind covalently to DNA. The covalent bond in turn leads to DNA damage.

Altretamine itself is a prodrug that is metabolized in the liver by oxidative N -demethylation to the actually active N-( hydroxymethyl ) melamines. The active ingredient is - taken orally - also because of its extremely poor solubility. Poor bioavailability in the oral administration is one of the main drawbacks of the compound.

Altretamine is usually in 14 - administered to 21 -day intervals between the cycles - to 21 - day cycles with 14. The side effects when taking basically are: loss of appetite, nausea and vomiting, and a general feeling of weakness.

Altretamine is weakly mutagenic.

The application of Altretamine is widespread common in the U.S. than in Germany. In Germany, Austria and Switzerland, and also various other European countries Altretamine - containing finished product are not or no longer on the market. Altretamine ( orphan drugs) is reported in the U.S. as drugs for rare diseases.
