On September 19, 1650 Astalli was appointed cardinal by Pope Innocent X and was Kardinalnepot, as his predecessor in office, Camillo Pamphili, married and the other predecessor in office, Francesco Maidalchini, had fallen out of favor with the Pope. On October 17, 1650, he was appointed Cardinal Priest of San Pietro in Montorio. When his mentor Panciroli died and Pope Innocent X died in 1655, he worried about his future in the Vatican and joined the Spanish group to Philip IV of Spain. In February 1654 he was released by Pope Innocent X and removed from his office due to state secret betrayal of Spain. Only after the death of Pope Innocent X, he came back to influence the Vatican. On January 24, 1661, he was elected by the cardinals to Camerlengo, and on July 4, 1661, he became Archbishop of Catania. Astalli took part in the conclave in 1655.
