Porcine ( pig liver esterase german, PLE) is a product obtained from the liver of pigs enzyme.

Description

Pig is obtained by extraction with organic solvents from pig livers. It is a slightly brownish powder, which can dissolve well in water.

The enzyme exists in three isoforms: The α - subunit has a molecular weight of 58.2 kDa, the β - subunit has the γ 59.7 kDa and 61.4 kDa subunits. The pig itself comprises three sub- units of all three isoforms. The main components of the natural product are ααα, ααγ, αγγ and γγγ. The different variants also have a different substrate specificity. The α - subunit was compared with the γ - subunit and a smaller proportion of aspartic acid for arginine at a higher. Since 2001 there has been next to the pig liver extract a porcine recombinant.

Use

The pig is the esterase used in synthetic chemistry at the most common and versatile. A series of reactions can be carried out stereoselectively using pig.

The enzyme may also be a polymeric support, such as polyacrylamide gels ( Eupergit ®), are immobilized. The activity of the enzyme is thereby degraded slightly and reaches about 68 % of the value of the non-immobilized pig.

Inhibitors

Trifluoromethyl ketones such as 1,1,1-trifluoro -4- phenylbutane -2-one is to inhibit the enzyme in the layer.
