Cyproheptadine (trade name: Peritol ®) is a drug which is used, among other things, for the treatment of cold urticaria.

It was patented in 1961 by Sharp and Dohme.

Clinical information

Areas of application (indications )

The use of cyproheptadine is hardly significant. The effectiveness of the drug for various demanding applications is poorly documented. The only drug law approved in Germany application for the treatment of cold urticaria cyproheptadine is when conventional antihistamines are not sufficiently effective. In the United States cyproheptadine preparations are also permitted as antiallergic agents for the treatment of seasonal allergic rhinitis or vasomotor, allergic conjunctivitis and allergic skin manifestations, for the relief of allergic reactions to blood or plasma as well as for the treatment of dermographism. In veterinary medicine Cyproheptadine is used to stimulate the appetite, but the drug is not approved for animals.

Outside the admission cyproheptadine is used to treat serotonergic syndrome, a disease which is mostly caused by overdosing or interactions of serotonergic drugs recommended. An effectiveness of cyproheptadine in the treatment of post-traumatic stress disorder, however, could not be proven. In the preventive treatment (prophylaxis) of migraine cyproheptadine is considered middle of the third choice. Its effectiveness is hardly occupied for this field of application.

Pharmacological properties

Is at cyproheptadine a tricyclic compound which is an antagonist at histamine H1 receptors and serotonin receptors, and has a weak anticholinergic effects. Experimental cyproheptadine inhibits also the acid sphingomyelinase.
