Lactacystin is an organic compound which is synthesized by the bacteria of the genus Streptomyces and has been described in 1991 first.

Effect

Lactacystin is a proteasome inhibitor and inhibits specific catalytic subunits of the proteasome. Proteasome is an enzyme that degrades intracellular proteins and one of the peptidases. Thus, it participates in the regulation of the cell cycle. The control of the Proteasomfunktion therefore potentially suitable for cancer therapy. Lactacystin was the first non -peptidic proteasome inhibitor, and is now widely used in biochemistry and cell biology.

Synthesis

The first total synthesis of lactacystin was developed in 1992 by EJ Corey and later supplemented by an efficient and direct enantioselective synthesis. Meanwhile, several research groups have developed alternative syntheses.
