Phosphate buffered saline

Phosphate buffered saline ( PBS short of English phosphate buffered saline) is a buffer solution that is used in biochemistry. The solution contains 137 mM sodium chloride, 2.7 mM potassium chloride and 12 mM total phosphate ( in the form of HPO42 - and H2PO4 - ). The pH of the buffering solution is 7.4. The property as a buffer solution allows you to work with this constant pH. The various salts, the solution having osmotic pressure of human body ( isotonic saline solution).

Application

PBS has many applications, since the solution is not isotonic and harmful to cells. It can be used to dilute substances to purify cell cultures or used to extend the life of immobilized biomolecules extension, such as proteins or enzymes. Additive agents such as EDTA may be added.

Composition

One liter of solution contains:

  • 8.0 g of sodium chloride (NaCl)
  • 0.2 g of potassium chloride (KCl)
  • 1.42 g of disodium hydrogen phosphate ( Na2HPO4 )
  • 0.27 g of potassium dihydrogen phosphate ( KH2PO4 )
