Positional cloning

The Positional cloning includes methods for identification of unknown DNA sequences in the immediate vicinity of a known DNA sequence. In contrast to the functional cloning the positional cloning is based on the search of an adjacent DNA sequence on the same DNA molecule. These adjacent DNA sequence possibly belongs to the same operon as the known sequence. The DNA is purified by DNA isolation and primer walking is determined ( a method of DNA sequencing ).
