Celebrit or Glucosulfon is a drug with Sulfonstruktur which has been investigated for the treatment of malaria, tuberculosis and leprosy. It is a prodrug that is metabolized to the therapeutically effective form of dapsone.

History

The first synthesis Promins is sometimes attributed to Edward Tillitson and BF Tuller of Parke, Davis & Co. drug in August 1937. In fact, Emil Fromm together with J. Wittmann synthesized various sulfone in 1908, among others, dapsone and some of its derivatives, as well celebrit. Fromm and Wittmann but devoted to chemical tests, not for drug discovery; Therefore, for decades the medical applications of these compounds have not been studied.

The unexpected discovery of the value of synthetic compounds such as sulfonamides in the treatment of microbial diseases, the scientists brought forward to investigating sulfones as a medicinal product. Earlier studies have shown disappointing results, but later celebrit and dapsone proved their value in the treatment of mycobacterial diseases. These sulfones were the first drugs that were hoping to get such inflammation under control.

First celebrit seemed to be less risky than dapsone; therefore, it was further examined at the Mayo Clinic in a guinea pig model as a means of treatment for tuberculosis. Because it was known that leprosy and tuberculosis caused by mycobacteria ( Mycobacterium leprae and Mycobacterium tuberculosis ), Guy Henry Faget asked by the national leprosarium in Carville, Louisiana, the company Parke- Davis for information on dapsone. Parke- Davis informed him further over the studies of leprosy in rats that had been carried out by Edmund Cowdry at the Medical School of Washington University in St. Louis. His 1941 published successful results convinced Faget with investigations to begin on humans; with celebrit and sulfoxone sodium, a related substance which had been developed by Abbott Laboratories. The first experiments were performed with six volunteers and later repeated in different places with larger experimental groups. Despite severe side effects, which led to the first attempts were briefly interrupted, you came to the conclusion that promin was effective. This breakthrough has been published worldwide, and reduced the fear of leprosy, and therefore also led to better treatment of patients who were still referred to at the time as " inmates", and could not use public transportation.

Pharmacology

And is water-soluble and can be administered intravenously Promin. Due to the heat stability of Promin injection preparations can be sterilized by heating.

Despite its water solubility celebrit has compared to the orally active dapsone can not enforce. There is no substitute for this even in case of intolerance to dapsone, since such intolerance is a group reactions to sulfone general.

Today, the drug of choice for treatment of leprosy dapsone, rifampicin and clofazimine.
