Thiamine diphosphokinase

Thiaminpyrophosphokinase ( TPK) is the enzyme that catalyzes the conversion of thiamine (vitamin B1) thiamine pyrophosphate (TPP). This phosphorylation is necessary as thiamine itself has no use in metabolism. TPK is found in all eukaryotes, in man, it is especially in the heart, kidney, testis, small intestine and leukocytes localized.
Variants of the enzyme may be associated with differences in birth weight. The regulation of TPK via the Sp1 cis- element.

Thiamine is phosphorylated to TPP. As a cofactor magnesium is necessary. Also Pyrithiamin is accepted as a substrate.
