Thromboxane-A synthase

Thromboxane synthase ( txas ) is the enzyme which catalyses the rearrangement of prostaglandin H2 to thromboxane A2. Txas is produced in chordates. In humans is the enzyme in platelets, of the lungs, the kidneys, can be found in the spleen and macrophages. The reaction product thromboxane promotes the narrowing of blood vessels and is the natural antagonist of prostacyclin. Mutations at TBXAS1 gene can lead to txas and Thromboxanmangel, as well as a ( rare ) dysplasia.

Txas target is in the treatment of chronic inflammatory bowel disease, as well as for bladder cancer treatment to reduce the amount of the chemotherapeutic agents used. Even with colon cancer and prostate cancer txas production is increased.

Catalyzed reaction

Prostaglandin H2 is rearranged to form thromboxane A2. The reaction is self-regulating.
