Cycles, or 1,4,7,10 -tetraazacyclododecane, is a cyclic polyamine. Cyclen is the starting material for DOTA and DOTA derivatives which have a wide application as drugs.
Synthesis
Compounds can be prepared by the combination of two nucleophilic substitution reactions. With the synthesis shown below, first described in 1974 by Richman and Atkins can also be produced many related macrocyclic Polyazaalkane.
Use
Cycles has excellent complexing properties. Thus cations are bound similar to crown ethers selectively. A large part of the cyclene is technically processed and reacted with bromoacetic acid to DOTA or DOTA - derivatives obtained. As the nitrogen atoms, in contrast to the oxygen atoms of the crown ether will allow an additional bond, similar to the DOTA, also (multiple ) coupled to receive more residues, such as adenosine, with new properties. This very strong complexing agents are widely used in diagnostics as well as in therapy.
