Functional cloning

Functional cloning includes methods for identification of unknown variants of a known gene, for example from other species or different isoforms or homologous genes within an organism. In contrast to the positional cloning of the functional cloning is based on the search of a gene similar DNA sequence and therefore presumably similar function.

Properties

The DNA is purified by DNA isolation, and then fragmented. By agarose gel electrophoresis, the DNA is separated and transferred by Southern blotting on a membrane. By hybridization with a hybridization probe, which is based in their DNA sequence to the known gene -like sequences on the membrane can be bonded. The sequences of binding DNA fragments are determined by DNA sequencing.
