The R6 begins at the outer ring expressway from Prague Rychlostní silnice 1 at the Prague airport.

The section between Karlovy Vary ( exit 131 - Jenišov ) and Cheb (exit 169 - Cheb -sever ) is highway toll since 19 April, 2012.
